I have an old unused iPod classic 60GB and can still fully charge the battery. I want to test it to find out how many hours of playing time it will give me and in the test will it matter if I have head phones attached? Thanks

It basically should matter just a bit if they are plugged in or not. There is certain small power consumption which is used to output the sound to the headphones, but not sure on how much difference it will make in battery lifetime. For a daily life scenario you better keep them plugged in, that way you will notice when the iPod battery will be empty - I'd say a positive side effect 
I think it will matter a lot for a real life test if yoi turn on dispaly all 10 minutes to switch tracks. No one is listening to one song opr album forever and the devices are shut down, started, then palylist changed and dfisplay usually is draining power more than just the headphones or something else....
