Skype video is not yet supported on the Mac. I have read somewhere that video support for Skype on the Mac "is very close", whatever that means. The version of Skype for the Mac is a few numbers behind the PC's version.
Video support for Skype on the PC is also still only in beta, to the best of my knowledge, so it will likely take a little while before it's all done.
What I can say about Skype on the Mac (I have an iMac), is that I use my cellphone's Bluetooth headset paired up with my Mac, and I walk hands-free chatting via Skype. Very cool.
As for PC-to-Mac video, check this out: Mercury Video

The answer to your question is yes. Windows supported programs made for mac with webcam support do accept iSight. But not all of them. For example, Yahoo just a couple days ago released a new version of yahoo messenger for mac which now supports webcams, including iSight. I haven't tried any other programs with it.
