Yesterday I bought an external hard drive so that I could put all my iMovie documents on it--I was done to 3GB on my HD and down to 4GB on a flash drive, and my latest movie just did not run smoothly: the music played, but the flow of the clips was halting. Once I had the EHD mounted, here are the steps that I took and the problems I faced:
1. I started copying the entire iTunes library onto the EHD; when I realized that the copy alert's indication that the process would take 7 hours or so to transfer nearly 8GB of data was accurate, I stopped the process.
2. I selected the folders already copied from the HD and trashed them; I then created a separate folder for music that I would need right away, and began to copy again.
3. Again the copy alert indicated that the process would take several hours, again I interrupted the copying, selected folders already copied and trashed them; after several tries, I finally got a selection which the program could copy in under half an hour.
4. I decided to try to copy the iTunes player onto the EHD; a pop-up alert informed me that not all of the data would transfer, and asked me whether I wished to proceed anyway. I clicked yes.
5. I tried to copy one of my iMovies onto the EHD; a pop-up alert informed me that a file icon could not be copied, and asked me whether I wished to continue. I clicked yes.
6. The copying process did not continue. I decided to copy the audio files, the media files, the imovie project original, and the QT movie in separate batches. This seemed to go well.
7. When, however, I opened the QT movie, the images played, but not the music.
8. When I attempted to open the imovie project original, a pop-up informed me that two files were missing, so iM could not open; I checked the files--both files were in the folders.
9. When I opened iTunes, the Library window was completely blank. Twice in the last two years I have found the iTunes library window unaccountably empty; once all the files disappeared, so I had to reconstitute the entire library; the second time, the files remained, but I had to call them up one by one back into the library. This time, when I double-clicked a file, the song would appear in the library window, and all signs indicated it to be playing, but I heard nothing.
10. I tried iTunes on both the HD and the EHD; silence.
11. I decided to download iTunes again from the Apple site; but once again, when I double-click the file icon of a song, it appears in the window and iTunes indicates it to be playing, but silence reigns.
12. When I open iMovie, more silence still, even though, when I select the first audio track, the edit volume guage reads 129%.
I don't know if I have one single problem affecting all three programs, or three different problems. When I tried to download iTunes from Apple directly onto the EHD, a pop-up window informed me that this could not be done. I imagine that, whatever I did to transfer a portion of the iTunes program to the EHD after the warning involved me in a dire mistake.
I'm sorry to be so long-winded, but I hope I've been careful enough for knowledgeable persons to see more or less what I, in my ignorance, have done wrong. I'd really appreciate any help!
Thanks!

Just to cover another base, launch Audio MIDI Setup.app in your
Utilities folder. What does it tell you about the Audio Devices? The
default output and system output should say Built-In Audio. The
formats should be 44100.0Hz, 2Ch-16Bit.
Take a look and get back to us.

Dear albloom,
I opened Audio MIDI, and all checks out.
I discovered just a while ago that there's no sound when I start streaming video on-line, either, so I imagine the problem is more general.
Thanks for the quick reply. I appreciate your willingness to be patient with me and to take time away from the holidays to help me out.

Not sure if I missed it but you only mention these programs; 1.do you have sound anywhere else (alerts, music played through other programs, besides the streaming which could be QT related) ?
2. And you have checked sound level hasn't been turned somewhere in programs and/or OS X (iTunes has a level f ex)?
